For FLT, please read this document: http://www.m17n.org/m17n-lib/m17n-docs/m17nDBFormat.html#mdbFLT and the other FLT files (e.g. DEV-OTF.flt, MLM-OTF.flt).
If you have in mind specific fonts and they are free, please send them to us. We'll develop the first version of FLT, and you can tune it up.

The m17n library doesn't provide a locale but just utilizes a locale supported by the system.
As for Dzongkha keyboard, I've just found this page:
